The role of Host Genetics in susceptibility to severe viral infections in humans and INSIGHTS into host genetics of severe COVID-19: A systematic review
Background: The vulnerability to a severe viral infection reported to be associated with genetic variants in immune response genes using case reports and studies GWAS. SARS-CoV-2 is a viral disease that appears that caused millions of COVID 19 cases worldwide. Approximately 15% of severe cases and some of them are accompanied by an immune system that is not regulated and the cytokine storm. There is increasing evidence that severe manifestations COVID-19 may be associated with human genetic variants in genes associated with immune deficiency and or inflammasome activation (cytokine storm).
Objective: Identification of candidate genes that may be helpful in explaining the severe COVID-19 and provide insight to understand COVID-19 is severe.
Methods: In this article, we systematically genes associated with susceptibility virus reported in human genetic studies (GWAS case reports) to understand the role of the host virus interactions and to provide insight into the pathogenesis of severe COVID 19th.
Results: We found 40 genes associated with susceptibility virus and 21 of them were related to SARS-CoV-severe and 19 severe COVID. Some of those genes involved in TLR pathways, other people in line C-lectins, and others associated with inflammasome activation (cytokine storm).
Conclusion: This compilation is a list of candidate genes that may be helpful in explaining the proper COVID-19 gene panel and included in the meta-analysis for different variants in genetic studies of a number of severe COVID 19th. In addition, we provide several hypotheses for the 19th COVID severe and possible therapeutic targets.
Evidence for non-genetic inheritance of germline human phenotype and disease
It is becoming increasingly clear that certain inherited across generations phenotype independent of the information contained in the DNA sequence, by factors in the germ cells remain largely uncharacterized. As evidence for the inheritance of non-germline genetic phenotype and disease continues to evolve in a model organism, there are fewer reports of this phenomenon in humans, because of various complications in evaluating the legacy of this mechanism in humans. This review summarizes evidence-based legacy non-germline genetic in humans, as well as significant challenges and important warnings that should be considered when evaluating this process in the human population.

Most reports of this process of evaluating the relationship of lifetime exposure to ancestors with DNA methylation changes or small RNA expression in germ cells, as well as the relationship between the experience of ancestors and inheritance phenotype in offspring, down to the great-grandchildren in some cases.
